What to expect from the Transgenic Mouse and Gene Targeting Core

Transgenics

• Emory Core will purify plasmid construct: BAC constructs should be purified by PI

• Emory Core provides pronuclear injection into either FVB/N or C57BL/6 strains

  • FVB/N-Plasmid or BAC constructs

  • C57BL/6-Plasmid constructs only

• Emory Core will send tail biopsies for genotyping anaylsis

• Emory Core guarantees two founder lines

Gene Targeting

• ES Cell Targeting by Emory Core

  • Standard ES cell line : HZ2.2 (129SV/Ev.TAC)

  • ES Cell targeting/expansion of HR clones
    (NOTE: Identification of HR targeted clones is not guaranteed)

  • Core provides DNA from 200 drug resistant clones

  • (optional) - ES Cell cassette deletion in-vitro

• Production of Knock-out/Knock-in mice

  • Germline transmission is guaranteed for ES cell projects performed by the Core

  • Germline transmission is not guaranteed for ES cells targeted by the PI
